Description

Poly(A) Polymerase uses ATP as a substrate for template-independent addition of AMP from ATP to the 3´ hydroxyl termini of RNA molecules.

Applications
  • Addition of a poly(A) tail to RNA synthesized in vitro for increasing RNA stability and translation efficiency prior to transferring into eukaryotic cells

  • cDNA library construction using a primer containing poly(dT) on its 3´ end.

  • Synthesis of polyadenylated RNA for nucleic acid amplification

Unit Definition 1 unit is defined as the amount of enzyme that will incorporate 1 nmol of ATP into acid-insoluble material in 10 minutes at 37°C.
Components
  • Poly(A) Polymerase: 5,000 U/ml in 20 mM Tris-HCl, 300 mM NaCl, 1 mM DTT, 1 mM EDTA, 50% Glycerol, 0.1% Triton® X-100, pH 7.5 @ 25°C
  • 10X Reaction Buffer: 500 mM Tris-HCl, 2.5 M NaCl, 100 mM MgCl2, pH 7.9 @ 25°C
Quality Control The absence of endonuclease, nicking activity, exonuclease and RNAse activity is confirmed by appropriate quality tests.
