Description

Pfu DNA Polymerase is a thermostable DNA polymerase isolated from an E. coli strain that carries the pol gene from Pyrococcus furiosus. It is one of the most thermostable DNA polymerases known to have 3’ to 5’ proofreading activity, the enzyme catalyzes DNA systhesis at optimal temperature near 75°C with very low error rate.

Unit Definition One unit is defined as the amount of enzyme required to catalyze the incorporation of 10 nmol of dNTPs into acid insoluble material in 30 minutes at 74°C under standard DNA polymerase assay conditions.
Components
  • Pfu DNA polymerase: 5 units/µL in 20mM Tris-HC1, pH 8.2, 1mM DTT, 0.1mM EDTA, 100mM KC1, 0.1% Nonidet P40, 0.1% Tween 20, 50% glycerol and enzyme stabilizers.
  • 10X Reaction Buffer: 200mM Tris-HCl (pH 8.8 at 25°C), 100mM (NH4)2SO4, 100mM KC1, 1% Triton X-100, 1mg/ml BSA, 20mM MgSO4
